Molecular Cloning of the Human Genes(s) Directing the Synthesis of Nervous System Cholinesterases.

Abstract

Complementary DNA (cDMA) and genomic DNA clones coding for human cholinesterase (ChE) were selected from phage DNA libraries by synthetic oligodeoxynucleotides coding for the organophosphate binding site of ChE and were characterized, sequenced and expressed within bacterial cells to yield AChE-like proteins. The Xenopus oocytes microinjection bioassay for translation of ChEmRNA has been improved and refined for studying the post-translational processing of nascent ChE molecules. ChE polymorphism and auto-immune anti-ChE antibodies were studied. Keywords: Acetylcholinesterase; Pseudocholinesterase; Immunoblotting; Poisoning; Cholinesterase inhibiters.
