Isolation of Genomic Clone for Human Cholinesterase.
Abstract
The goal of this work is to isolate the gene for human serum cholinesterase (EC 3.1.1.8). We have succeeded in isolating a partial clone that codes for amino acids phenylalanine 195 through leucine 574, and contains 302 bp of the 3' untranslated region. This clone was found by screening a human brain Complementary Deoxyribonucleic Acid (cDNA) library with oligonucleotide probes. The probes corresponded to the amino sequence of human serum cholinesterase. The coding region of our 1440 bp clone matched the amino acid sequence of human serum cholinesterase perfectly. This suggested that human cholinesterase was identical in brain and in serum. We are continuing to search the brain cDNA library for other cDNA clones that will extend our present clone in the 5' direction so as to have the complete coding region of cholinesterase.
