Evaluation of Diethyleneglycoldinitrate (DEGDN) and Two DEGDN-Containing Compounds

Abstract

This study evaluates diethyleneglycoldinitrate (DEGDN) and tw o DEGDN-containing compounds, DIGL-RP and JA-2, for mutagenicity and carcinogenicity in mammalian cells in vitro. The potential mutagenicity of the three test articles was assessed by point mutation at the thymidine kinase locus in the mouse lymphoma cell (L5178Y TK+/-) forward mutation assay. The potential carcinogenicity of the three test articles was assessed by in vitro cell transformation as measured by anchorage-independent growth of Rauscher leukemia virus infected rat embryos cell (RLV-RE). The results indicated that the three test articles contained direct-acting mutagen(s) which was shown by increased mutagenic activity without the addition of rat liver S-9 and the lack of enhanced mutagenic activity with the addition of the rat liver S-9 activation. Linear dose-mutation responses of statistical significance were detected with DEGDN, DIGL-RP and JA-2; DEGDN had the effect of a weak mutagen while DIGL-RP and JA-2 had the effects of a strong mutagen.
