Vector Competence of Mosquitoes for Arboviruses

Abstract

Aedes aegypti mosquitos are very refractory to peroral infection by dengue 2 (New Guinea C) virus; ID50's in excess of 7 Log10 PFU/ml ingested. Parenteral infection ID50's are slightly above 2 Log10 PFU/ml. Binding of western equine encephalomyelitis (WEE) virus to brush border fragments (BBF) from mesenteronal epithelial cells of susceptible (WS) Culex tarsalis was greatly in excess of that to refractory (WR) Cx, tarsalis and Culex pipiens. The binding was specific. There are an estimated 1.7-3.4 x 10 to the 6th power binding sites/mesenteronal epithelial cell and the affinity constant is 2.3 x 10 to the 11th power/M. Monoclonal antibodies were produced against the BBF of mesenteronal epithelial cells. Three hybridomas (13A5, 13C1, and 20C5) were subcloned and ascites fluid produced. The 13A5 and 20C5 hybridomas are IgG2b subtypes and 13C1 is an IgM. Further biochemical analysis of BBF proteins revealed that Cx. pipiens closely resembles WR Cx. tarsalis in its 2D electrophoretic pattern. Reversed phase high performance liquid chromatography did not separate the major proteins of interest. A congenic line of high WEE virus producing (HVP) Cx. tarsalis was derived from low WEE virus producing (LVP) Cx. tarsalis after 13 generations of backcrossing.
