Development and Validation of Methods for Applying Pharmacokinetic Data in Risk Assessment. Volume 6. Sensitivity and Uncertainty Analysis

Abstract

Model parameter uncertainty/sensitivity analysis was addressed for the use of physiologically-based pharmacokinetic (PBPK) models in risk assessment applications. Both uncertainty in input parameters and the corresponds effect on model output was examined. An assessment was made for the individual model parameters as they relate to the overall uncertainty of model output. This volume describes a sensitivity analysis conducted using a relatively simple PBPK model. Parameter values were varied one at a time by an arbitrary, small percentage. Also presented are the preliminary considerations and experimental data relevant to the conduct of an uncertainty analysis.
