Suicide Inhibitors of Reverse Transcriptase in the Therapy of AIDS and Other Retroviruses

Abstract

A rapid assay procedure for the enzyme has been developed based upon dot blotting acid washing using DE filter paper dises. Experimental parameters in the procedure have been carefully defined and controlled so that the assay is of sufficient precision for enzyme kinetics. The procedure is also used as a routine assay of reverse transcriptase in virus supernatants thus providing a quantitative measure of virus replication in tissue culture. A comprehensive study of the template and substrate dependence for inhibition of reverse transcriptase by Foscarnet has been completed and published as detailed in this report.
