Structure and Expression of Genes for Flavivirus Immunogens.

Abstract

Progress during the report period included: (1) extension of the DEN- 1 sequence analysis; (2) napping of binding sites for anti-DEN-l/JE monoclonal antibodies; (3) development of nonradioactive probes for 'clinical diagnosis of Dengue; (4) assembly of large clones of DEN-1 cDNA; (5) optimization of procedure for transfection of cells with DEN-1 RNA; (6) investigated ratio between plus- and minus-strand dengue RNA in infected cells; (7) expressed fusion and non-fusion derivatives of DEN-1 proteins in bacterial and eukaryotic systems, including expression via recombinant vaccinia and baculoviruses. Neutralizing antibodies were elicited by 'E. coli-expressed antigens; neutralizing and protective antibodies were raised in mice against vaccinia-JE recombinants.
