Vasopressin Receptor Signaling and Cycling of Water Channels in Renal Epithelia
Abstract
Water reabsorption and urinary volume regulation are important processes in maintaining normal physiological function. Vasopressin (ADH) is the hormone that is responsible for regulating water homeostasis. We have been examining the mechanism whereby ADH enhances water permeability in renal epithelia. Our observations suggest that an integral component of the water reabsorptive process is the enzyme, protein kinase C (PKC). Using immunocytochemical techniques and enzyme assay procedures, we have identified PKC isozymes that translocate from the cytosol to the membrane during activation by ADH and PKC activators. We also report on an in vitro model of cultured kidney cells for measuring transepithelial fluid and electrolyte transport and have evaluated various filter supports for their transport characteristic limitations.
