Functional Properties of Nonhuman Primate Antibody to Prophyromonas Gingivalis

Abstract

The nonhuman primate (NhP) serves as a useful model for examining the host-parasite interactions in Pg-associated periodontal disease. This study determined the influence of NhP sera on: (1) direct killing of Pg 3079.03, (2) Pg induced superoxide anion (O2-) release from human PMNs; and (3) the ability of PMNs to bind and phagocytize Pg. Three types of NhP sera were utilized: (1) normal/baseline (BAS), (2) obtained after ligature induced periodontitis (LIG); and (3) following active immunization with formal in killed Pg (IMM). Fluorescent dyes consisting of DAPI and propidium iodide were used to determine bacterial viability. Antibody mediated killing, and PMN Attachment/ingestion of Pg were assessed by mixing sera, and Pg, or Pg and PMNs, noting the percentage of viable/nonviable cells, and the ratio bound and phagocytized Pg to PMNs using fluorescent microscopy. O2- production was quantitated spectrophotometrically by cytochrome c reduction and was used as an indirect measure of neutrophil activation
