Three Dimensional Structure Determination of Botulinum Neurotoxin

Abstract

To determine the 3-dimensional structure of botulinum neurotoxins and their isolated domains. The following specifications are listed in the contract section C-Statement of Work: Crystallization of the 150 kDs holo-botulinum neurotoxins. Serotype A has been crystallized previously and crystallization conditions will be refined as necessary. Serotype B will be crystallized. Determination of the 3-dimensional structure of serotype A. Heavy atom derivative screening is underway. Once 2 or 3 'acceptable' derivatives are known, data collection and processing of native and derivative data will be completed. The phases of the x-ray diffraction pattern will be phased, electron density maps calculated, and the structure will be determined
