Tissue Banks and Cell Lines Derived from Patients with a High Risk of Breast Cancer and In Situ Disease.
Abstract
This project is aimed at establishing bulk cultures of individual epithelial and stromal cells from mastectomy specimens of patients who are carrying genes that predispose them to breast cancer. Epithelial and stromal cells are being conditionally immortalized using a novel amphotropic retroviral vector transducing the tsA58-U19 SV40-derived construct. These cells are from patients who are mutation carriers of BRCA1, BRCA2, and p53. Samples have been received from 38 women, eight of which have a Li-Fraumeni - like phenotype, four with confirmed mutations. Other cases are presumptive carriers of BRCAl, BRCA2 or other mutations. In two cases the mutations have been identified and the others will be screened in the next year. In the majority of cases primary cultures have been established, cells banked and immortalization commenced. In parallel a detailed data-base is being established of in situ disease from patients that have been sent to the Royal Marsden Hospital Breast Unit since 1970. Over 5,500 slides have been reviewed of in situ breast cancer from over 700 cases. Material from over 500 patients has been selected for further study. The cases from 1994-96, with short follow up are being used to pilot the molecular studies. Sections have been cut and studies commenced on the molecular and immunocytochemical analyses of these lesions.
