Characterization of an In Vitro Human Breast Epithelial Organoid System

Abstract

We have studied the expression of estrogen receptor (ER) in two types of normal human breast epithelial cells (HBEC) and their SV4O transformed cell lines. The results show that Type I HBEC expressed a variant ER by Western blot analysis. This variant ER contains a deletion in the DNA binding domain (exon 2) as revealed by RT-PCR analysis. In contrast, Type II normal HBEC are ER-negative. SV40 transformed Type I and Type II HBEC also expressed this variant ER. Tumors formed in athymic nude mice by in vitro transformed Type I cell lines, however, expressed a high level of wild type ER. Similar differential ER mRNA splicing was observed between these cells grown on plastic and in Matrigel.
