Suppressor Genes in Breast Cancer

Abstract

The current proposal will focus on the isolation and characterization of novel TSG(s) in human breast cancer. A human epithelial eukaryotic cDNA expression library has been constructed, and will be transfected into the human breast cancer cell line MCF-7. Gene(s) inhibiting the growth of MCF-7 cells will be considered candidate TSGs for breast cancer. These will be cloned and the full length cDNA sequence obtained. Expression of cloned genes will first be investigated in RNA populations derived from two immortalized "normal" human breast epithelial cell lines (AlN4 and MCF-lOA), and in MCF-7 cells growth arrested either by antiestrogen- treatment or estrogen withdrawal.
