Regulation of Growth Factor Release By Proteases Inhibitors in Breast Cancer

Abstract

This report describes the cloning, sequencing and properties of a potential TGFa-releasing protease present in human breast epithelial cells. Degenerate oligonucleotides based on the conserved sequences about the his(57)and ser(195) residues in mammalian serine proteases were used to clone a novel serine protease sequence (PCR-7) from an MCF-7 breast cancer cell cDNA library. The deduced 321 amino acid open reading frame shows a high degree of homology with the cell-surface serine proteases enterokinase, prekalikrein, hepsin and prostasin. Northern blot analysis, using a PCR-7 cDNA probe, shows the presence of an approximately 4.2 kb mRNA species in total RNA from MCF-7 cells.
