Promoter Regions Determining Over-Expression of Metalloproteinase Genes in Breast Cancer

Abstract

The purpose of this project is to identify the molecular and genetic basis for the abnormal destruction of extracellular matrix that accompanies breast cancer, by addressing the transcription of the genes of the matrix metalloproteinases (MMPs) that are made at high levels in and around carcinomas. The objectives of this project are to identify regions of the promoters of three MMP genes that mediate their high level of expression in vivo, to determine the histological distribution of MMP-producing cancer cells in tumors, to determine for each MMP gene whether it has undergone amplification.
