Vacuolar-Type H+-ATPases in Human Breast Cancer

Abstract

The amino acid sequence of the ll5kD subunit of the Vacuolar-type ATPase proton pump has been analyzed and predicted to have six membrane-spanning domains. PCR primers have been designed and used to amplify DNA sequences coding for five of six targeted nonmembrane spanning domains. The amplified DNA sequences have been purified and ligated into the pGEX-2T expression vector for generation of GST fusion proteins,to be used to test the specificity of OSW-2, a monoclonal antibody targeting the 115kD subunit, and to be used to generate antibodies targeting the nontransmembrane spanning domains of the subunit.
