Role of Angiogenic Growth Factor Receptors, Tie 1 and Tie 2, in Determining the Metastatic Potential of Breast Cancer.
Abstract
Angiogenesis determines tumor progression and microvessel density correlates with metastatic potential of breast cancer. Expression of endothelial cell receptors, Tiel and Tie2, was described in areas of active vascular remodeling. There is no information on the role of Tie receptors in breast cancer progression. We selected breast cancer tissues and normal specimens and analyzed a fraction of these samples for Tie expression. We found differences in mRNA content in malignant and benign breast tumors. These exciting observations suggest that Tie receptors play an important role in angiogenesis associated with breast cancer progression. We also established that expression of Tiel and 2 in human endothelial cells is induced by cell-cell interaction and is highest in adult quiescent endothelium. We will establish whether the frequency and pattern of expression of Tie I and 2 in breast cancer differ from expression in adjacent tissue and normal breast tissue. Statistical correlation with tumor progression will be obtained following analysis of dinical data. These studies will determine whether Tiel and Tie2 are novel markers of angiogenic potential of breast cancer.
