A Murine Model of Genetic and Environmental Neurotoxicant Action

Abstract

The purpose of this project is to study environmental and genetic interactions in the etiology of Parkinson's disease. The scope of the project is to use transgenic mice and tissue culture methods to determine the role of alpha-synuclein, MPTP, and paraquat in damaging dopaminergic neurons. The major findings to date include the generation and characterization of transgenic lines of mice expressing alpha synuclein in catecholaminergic cell groups, their increased vulnerability to MPTP and their vulnerability to combined manner and paraquat. A construct to generate somatic mosaic animals has been created and tested. It will be used to create a second model of alpha synuclein over expression. Viral vectors overexpressing alpha synuclein have been created, tested and result in synuclein aggregates in neurons. The first year has been devoted to the generation of transgenic mice and the creation of cloning molecules and vectors. Significant development of resources and techniques was expected and accomplished in this year. Later years will result in production of data to address specific hypotheses.
