Molecular Analysis of the Cripto Growth Factor Receptor

Abstract

Cripto is a member of the EGF-CFC gene family and is involved in mammary tumorigenesis. My research project focus on identifying and cloning of CRIPTO receptors. In order to detect CRIPTO specific binding protein, a co-immunoprecipitation assay using AP-mCRIPTO as affinity reagent has been established. Using this assay, two protein bands, a major 65 Kd band and a weaker 63 Kd band, were detected as specifically binding to AP-mCRIPTO but not AP. These two proteins were also shown to be cell surface proteins, suggesting that they could be CRIPTO receptors. The purification and sequencing of these two proteins are underway.
