The Role(s) of Heparan Sulfate Proteoglycan(s) in the wnt-1 Signaling Pathway

Abstract

The main purpose of my research proposal is to understand the regulation of Wingless signaling by heparan sulfate proteoglycan (HSPG). I have previously cloned the Drosophila sugarless (sgl) and sulfateless (sfl) genes that encode enzymes required for the biosynthesis of heparan sulfate glycosaminoglycans. In this report, I have conducted further characterization of sfl gene, which is required for Wingless (Wg) signaling in both embryonic and wing imaginal disc development. I have also identified Dally, a GPI-linked Glypican, as the HSPG molecule involved in Wg signaling. loss of dally activity, both in the embryo and wing imaginal discs, generates phenotypes reminiscent of loss of Wg activity.
