Intracellular Single-Chain Antibody Directed Against Cyclin D1 Oncogene as a Knock-Out Strategy for Breast Cancer

Abstract

Over expression of the cyclin D1 oncoprotein is linked to the transformed phenotype in the context of carcinoma of the breast. On this basis, we endeavored to achieve functional knockout of this target employing intracellular single chain antibodies (intrabodies). This intervention accomplished down regulation at cyclin D1 with altered cell kinetics. In addition, this intervention accomplished tumor cell chemosentization. Intrabodies against cyclin D1 thus represent potential therapeutic agents for carcinoma of the breast.
