Functional Characterization of the Murine Homologue of PLAB, A Novel TGF-B Member, in Placental and Mammary Tissues

Abstract

The goal of this proposal was to generate and analyze mice mutant for the mPlab gene. The gene was successfully cloned and sequenced. The targeting vector for production of null embryonic stem cells was constructed. Embryonic Stem (ES) cells containing the conditionally null allele of mPlab were created. While testing these ES cells for correct homologous recombination events, we discovered that the mPlab gene had already been knocked out by another laboratory. Due to a lack of phenotype reported by the other paper, we terminated our mPlab experiments and have begun designing a different set of specific aims. Here we detail the work generated under the mPlab studies, and present mini-proposal of our new research testing the mouse as a model for primate early development. If the reviewing board feels funding can be continued on the new research will submit a full proposal at its request.
