The Protein Kinase RSK Family - Roles in Prostate Cancer

Abstract

The Ser/Thr protein kinase p90-kDa ribosomal S6 kinase (RSK) is an important downstream effector of MAPK but its roles in prostate cancer have not previously been examined. We have now discovered that RSK2 regulates the expression of PSA, an important diagnostic marker for prostate cancer. Furthermore, RSK regulates the growth of some human prostate cancer cell lines. Taken together with our observations that RSK levels are higher in 30% of human prostate tumors compared to normal prostate tissue suggest that RSK is an important drug target for prostate cancer. To further test this hypothesis we are developing prostate cell lines that will inducible overexpress RSK and transgenic mice that will overexpress RSK in the prostate. RSK-specific inhibitors, such as SL0101, which we recently discovered, should be useful as chemotherapeutic agents for the treatment of prostate cancer. The in vivo efficacy of SL0101 to inhibit the growth of prostate cancer is currently being evaluated using a PC-3 xenograft model in mice.
