Prediction of Aggressive Human Prostate Cancer by Cathepsin B
Abstract
The author spent considerable effort on several revisions and follow-up discussions with Mr. Peter Marshall and Dr. Julie Wilberding to obtain required approval of the protocols from the HSRRB (Log No. A-12517). The last revision of his protocol (see 10 Feb 2005 letter) met the requirements of the HSRRB (Log No. A-12517). He received an approval memo dated 13 Apr 2005 from Col. Laura R. Brosch, PhD, that was received around 20 Apr 2005. Although the grant was funded as of 1 Mar 2004, he could not conduct any studies on human subjects until 20 Apr 2005. Earlier, he had sought clarification (1 Mar 2004) of the University memo as to whether he could spend funds to hire personnel. When Mr. Barry Quast, an experienced scientist, declined to accept the position, this necessitated hiring new personnel and training them in prostate research. The author received approval for hiring and spending funds prior to the HSRRB approval (Log No. A-12517). He began using limited amounts of funds after the expiration of the first year (1 Mar 2004 to 28 Feb 2005) of funding. He recruited and hired new employees and trained them in the methods of processing prostatectomy and biopsy tissue samples and immunohistochemical Elisa assay techniques for cathepsin B and stefin A and other markers, including cytokeratin and prostate specific antigen (PSA). They also were taught laser capture microdissection techniques; methods of transferring images of reaction products directly from microscopic slides to a computer equipped with a Metamorph image analysis system; quantification of localization data; data collection, storage, and retrieval; patient confidentiality; good laboratory practices; and VHA Privacy Policy. In summary, these employees were trained to perform all the tasks required for the study. The author began the project as soon as he was given approval by the DQD-HSRRB in April 2005.
