Tissue Culture Studies of Host versus Graft Reaction.

Abstract

In accordance with the contract, studies directed to the preparation of purified leukocyte blastogenic factors have reached the following stage. By a combination of sucrose density gradient centrifugation (Spinco Model 14 ultracentrifuge, B-4 rotor, 5-20% surcorse, 18 hrs. running time) and DEAE-Sephadex A-50 chromatography (Gradient: 0.1M-1.0M Tris-HcL, pH 7.6, 50C) 2 blastogenically active fractions have been reproducibly isolated in a highly purified form. One of the active fractions has been freed (detectably) of all human serum proteins and obtained in a yield of 0.2-0.5 mg/gm CFM (dialysed and lyophilized). The other active fraction contains L2-macroglobulin as the only serum protein component detectably present and is obtained in yields of 2-3mg/gm CFM. It is active at the 300-500 microgram level.
