Propagation in Serum-Free Medium of Cell Cultures Useful for Vaccine Production.
Abstract
We continued to attempt to adapt several diploid cells in serum-reduced, serum-free media employing monolayer and shaker cultures in order to eventually use such cells for production of dengue virus vaccines. All cells but monkey lung cells (DBS-FRhL-2) were gradually weaned from media containing 10% to 2% fetal calf serum (FCS). The media for FRhL-2 cells was specifically designed for direct use. Media were supplemented with defined components of serum which were reduced or eliminated from serum when initially using 2% FCS. The best level of achievement obtained for the following cells were: human lung cells (WI-38 and IMR-90), 4 serial passes in Waymouth medium supplemented with 2% FCS; monkey lung cells (FRhL-2), recently serially passed 15 times in our highly modified F15 medium with 2% FCS; and Aedes albopictus clone C6/36 cells, 10 serial passages in monolayers using F15FCS5 and 19 passages in F15FCS2, 5 serial passages using shaker cultures with F15FCS5 and 3 serial passages in F15FCS2, 11 serial passages using spinner cultures in F15FCS10.
