Drug Development of the Antimalarial Agent Artemisinin: Total Synthesis, Analog Synthesis, and Structure-Activity Relationship Studies.

Abstract

In the past year we have synthesized over 20 analogs of artemisinin (qinghasosu, QHS), 16 of which were submitted for determination of in vitro antimalarial activity. The analogs may be partitioned into four major groups: (1) optically active, substituted QHS and dihydro-QHS analogs, (2) racemic 6,9-desmethyl QHS and a truncated congener, (3) racemic and optically active seco-analogs with various lactone substituents, and (4) racemic acyclic QHS analogs.
