Synthesis and Antiviral Evaluation of Pyrazofurin Analogues.

Abstract

Synthetic methods have been investigated towards 5'-amino-5'deoxypyrazofurin, 5'-deoxy-5'-phosphonylpyrazofurin, 5'-deoxy-5'-phosphonylmethylpyrazofurin, a 5'-deoxy phosphoramidite pyrazofurin derivative, 3'-deoxy-3'-fluoropyrazofurin, and 5'-nor hydroxymethyl pyrazofurin. Preparation of the latter compound has been completed and this derivative will be submitted shortly for antiviral analysis. Antiviral data has been obtained for the N-methyl- and N,N-dimethylamide derivatives of pyrazofurin and for 5'-deoxypyrazofurin and 5'-homopyrazofurin. Only the N-methyl amide analogue showed any activity (versus influenza B only), which was insufficient to warrant further study.
