Drug Evaluation in the Plasmodium Falciparum - Aotus Model

Abstract

The Panamanian Aotus - Plasmodium falciparum model was used to evaluate antimalarial drugs, singly and in combination. Azithromycin cured Smith/ RE strain infections. Tetrandrine (WR 149544) plus chloroquine reversed chloroquine resistance in vivo as indicated only by parasite clearance not infection cure. WR 99210 retained its antimalarial activity when co-administered with folinic acid, a combination that may ameliorate toxicity of WR 99210. Results of trials (still current) to induce resistance to WR 250417 and WR 169626 show that such resistance may have occurred by treatment with subcurative doses and parasite subinoculations. A chloroquine-resistant P. vivax strain was adapted to Panamanian Aotus; drug resistance was con-firmed. A chloroquine- sensitive P. vivax strain was not infective for unaltered Panamanian Aotus.
