Fiber Optic Biosensor for Early Prediction of Infection

Abstract

During Phase I, IA established the feasibility of developing a biosensor for rapidly detecting early-stage serum infection markers by demonstrating the performance of fiber sensors which are able to detect levels of IL-6 and IL-12 in buffered fetal calf serum in approximately 10 minutes with range and specificities comparable with those achieved using ELISA methods (0.25-10pg/ml). After calibration, acceptable sensor to sensor reproducibility was demonstrated and steps necessary for further improving reproducibility during Phase II have been identified. IA also developed and demonstrated a single fiber biosensor which can sequentially measure levels of both IL-6 and IL-12 in a single undiluted human serum sample. Finally, during Phase I, IA has developed a design for a hand-held instrument for use by minimally trained personnel. The instrument which will be further developed during Phase II work will provide a means for early detection of infection in battlefield casualties as well as in civilian hospital or clinic setting. This will reduce the frequency with which infection results in sepsis by facilitating early treatment of infections.
