Characterization of Uterine Derived Growth Inhibitor (UDGI): A Novel Growth Inhibitor of Estrogen Receptor Negative Breast Cancer Cells
Abstract
We report the cloning and characterization of a novel cDNA, 0KL38. OKL38 gene expression was induced by pregnancy and hormone associated with pregnancy such as hCG. OKL38 expression was low in breast cancer cell lines and barely detectable in DMBA-induced breast tumours. Transfection of human MCF-7 breast cancer cells with OKL38 cDNA lead to reduction in proliferation and tumour formation in nude mice. The data suggest that the OKL38 may play a crucial role in the growth regulation and differentiation of the breast epithelial cells during pregnancy and tumorigenesis. We also report that coadministration of ICI 182780 and testosterone enanthate caused mammary gland differentiation and reduced tumour incidence, number of tumour bearing and tumour size. The data may be useful in designing novel strategies for breast cancer prevention and/or treatment based on maximizing mammary epithelial cell differentiation.
