Cell-Cycle Related Events in the Dimethyl Sulfoxide-Induced Commitment of Hepatic Tumor Cells to Enhanced Liver Function,
Abstract
The present data indicate that DMSO perturbs the proliferative kinetics of mouse hepatoma cells in vitro. This reduced proliferative capacity and concomitant stimulation in albumin accumulation, as a consequence of exposure to DMSO, are associated with an increase in the percentage of 4C, G2-like, hepatocytes in the culture. This expanding 4C compartment may represent a subpopulation of hepatocytes arrested (or prolonged) in G2. Tsukada and Hirai have shown that albumin synthesis is restricted to the mid S to late G2 phase of the hepatoma cell cycle. The marked increase in albumin accumulation, as a function of DMSO concentration, in the mouse hepatoma cells used in the present study is consistent with this finding. DMSO-stimulated hepatic tumor cells, therefore, may provide a useful in vitro system for investigations into the control of gene expression during the hepatocyte cell cycle.
