DURIP-22 ACQUISITION OF ATOMIC FORCE MICROSCOPE FOR DYNAMIC TOPOGRAPHICAL AND MECHANICAL CHARACTERIZATION OF BIOMATERIALS
Abstract
We request the fund to purchase an atomic force microscope (AFM) capable of measuring the topography and the mechanical properties of the sample at sub-micron resolution to study (1) the dynamic changes in organelles; (2) the relation among loading condition, resulting mineralization and mechanical property mapping of bone-inspired materials; (3) the mechanical behaviors of DNA-based metamaterials, and (4) characterization of nano-engineered surfaces and materials for the systematic study of cell behavior in vitro. The four PIs have long-established collaborations. The AFM requested will feature the capability of nano-meter surface topography imaging and pico-Newtown (pN) surface force measurement, in air or in liquids, as well as of dynamic parameter adjustment to optimize the image quality. Currently, we do not have access in the campus to AFMs of comparable capabilities. The instrument-sharing logistics will be established by following the example of many other instruments currently shared among our four closely collaborating groups. All of the projects listed in this proposal are in areas of interests to DoD. The acquisition of the AFM will offer an orthogonal verification of the test results by other approaches originally proposed in the submitted and funded proposals, will increase the resolution of the material characterization, will facilitate fast design-validation cycles, will enhance the quality of the DoD-related research, and will allow us to answer new questions risen from the ongoing research. We also designed research-related training programs, which utilize the access to the AFM and the data generated by the AFM to enhance the learning experience of students.
