Genome-Wide Mapping (GWM) of Host Proteome:Pan Alphavirus Proteome Interactions for Discovery of Broad-Spectrum Countermeasures

Abstract

Venezuelan equine encephalitis virus (VEEV), eastern equine encephalitis virus (EEEV), and western equine encephalitis virus (WEEV) are encephalitic alphaviruses that are highly infectious as aerosols and pose an important threat to the warfighter. We have successfully utilized discovery-based proteomics platforms including mass spectrometry (MS) to identify, validate and describe the mechanisms by which VEEV proteins interact with host proteins. ATCC proposes to extend these studies to structural and nonstructural proteins of VEEV, EEEV and WEEV and successfully identify inhibitors that disrupt these interactions, and hence can be utilized in a broad-spectrum manner to develop anti-alphavirus therapeutics.
