Optimizing DNA/EP-Based Antibody Gene Transfer Platform Technology

Abstract

The objective is to optimize antibody gene transfer conditions such that clinically relevant levels of serum antibody concentrations will be achieved in larger animal species. The investigators will use modeling and numerical simulations to guide optimizations of electroporation parameters specifically for the purpose of antibody gene transfer. The model will be tested experimentally, and the results will be used to further refine the model. Iterative experiments will be performed in rodents to develop the model followed by testing in rabbits and sheep to verify scalability. They will identify at least one optimized DNA vector candidate capable of producing sustained monoclonal antibody levels in mice and an electroporation protocol optimized for increased antibody expression in larger animals.
