Metastic Progression of Breast Cancer by Allelic Loss on Chromosome 18q21
Abstract
Mutations in the Smad2 and Smad4 genes, localized to chromosome 18q, a region frequently deleted in advanced cancers is rare in breast cancer unlike pancreatic, colon, lung, and ovarian cancers. However, the fact that 18q loss has been predominantly associated with the advanced stage of cancers suggests that the genes inactivated by this specific alteration or other genes in the same pathway targeted for the inactivation could be associated with the conversion of benign tumors to malignancy and metastatic progression of breast cancer. Our analysis showed that 30% of the breast cancers exhibit loss of Smad8 expression, making it on of the highly valued markers similar to Her/neu.
