New Drugs for Pretreatment of Organophosphonate Intoxication
Abstract
Our work has focused on the synthesis of compounds having the general structures shown below. Eighty-four compounds were synthesized and submitted to the U.S. Army Medical Research and Development Command (MRDC). In addition, two previously submitted 1((2-phenylmethyl)cycloalkyl)amines were resynthesized at the request of the Contracting Officers Representative (COR) for further in vivo testing. Forty-five 1-((1-arylcycloalkyl)methyl)amines were submitted. Structural variables examined included the phenyl substituent (unsubstituted, 3- OCH3, 3-OH, 3,4-(OCH3)2, 4-CH3, and 4-NO2), the cycloalkyl ring (cyclohexyl, cyclopentyl, cyclopropyl, and replacement of the cycloalkyl ring with two ethyl groups), the amine (pyrrolidine, piperdine, morpholine, diethylamine, and dimethylamine), the aryl group (substituted and unsubstituted phenyl, 2-thienyl, and 1-naphthyl), and protic verses quaternary (methyl) salts. Keywords: Pretreatment, Antidote, Organophosphonate, Synthesis, Acetylcholinesterase, Anticholinesterase.
